I?ve never played a Ben 10 game before and the only true knowledge I have of the series is that it had a very popular iteration a few years back that catapulted the franchise into a top seller. So going into Ben 10 Galactic Racing, I?had a completely open mind and was?ready for anything. Well, what I was given was a basic kart racer with graphics that would be underwhelming on the PlayStation Portable, let alone the PlayStation Vita.
Seriously,?if you stacked?Ben 10 Galactic Racing?up against Mario Kart DS, you would be?hard pressed to find a clear winner in the graphical department. The characters are bland, the?vehicles you race in have very little detail, and the environments are all dark and muddy. Having a game that looks this bad is heart breaking on my new 5??OLED screen.
After the shock of how?disappointing the graphics are wears off, you will be given a very miniscule amount of tracks that can all be unlocked within an hour. Although 25 tracks may seem like enough, they are all short and?many of them will overlap each other. You?ll play through the first few tracks in the Galactic Grand Prix, then later tracks just add onto them by about 20 seconds of new area. The level design is just so uninspired that it hurts,?and each?one becomes boring to?race in after a very short period of time.
When it comes to the actual kart racing, it is very standard fare. If you?ve played Mario Kart or Modnation Racers, you know exactly what you?re getting into, minus the charm. Sure, the game does feature power-ups but they all feel like carbon copies of those seen in other games in the genre. I know there isn?t much to add that Mario Kart hasn?t done before, but when you look at Modnation Racers, it had just enough wackiness to make the weapons feels fresh.
After you pass the very short Grand Prix, you?ll only be about 25% done the game but that doesn?t mean there is a lot to do. You can play the game in multiplayer with a few friends, or against the CPU in basic races?and arena battles. Other than that, if you want to unlock all of the trophies, you have to compete in the?Time Trials. The annoying thing about them is that?there is no set time to beat, so you have to finish a track, get your time, re-race the track, beat that time, and your trophy will unlock. Basically, you won?t?have?much?to do in the game after you beat the Grand Prix.?

An undated photograph from a Russian television report shows a man said to be Adam Osmayev, one of the suspected militants conspiring to kill Vladimir Putin. Both of Osmayev's hands were bandaged, and his face was covered in green dots from an antiseptic used to treat his cuts.

Updated at 9:25 a.m. ET: MOSCOW -- Security forces have uncovered a plot to assassinate Russia's Vladimir Putin and have arrested suspects linked to a Chechen rebel leader known for other terror attacks, Russian state television reported Monday.
Pro-government Channel One said that the suspects were plotting to kill Putin in Moscow immediately after the March 4 presidential election, in which he is all but certain to reclaim the presidency.
The report, which included televised confessions by two suspects, is likely to boost support for Putin as he seeks his third term as president in an election Sunday.
Channel One said the suspects were acting on instructions from Chechen warlord Doku Umarov and had been arrested in Ukraine's Black Sea port city of Odessa after an accidental explosion Jan. 4 while they were trying to manufacture explosives at a rented apartment.
Amanda Walker, Moscow correspondent for Britain's Sky News, pointed out that Channel One was a "staunch Putin supporter."
The Ukrainian Security Service said earlier this month it had detained a man sought by Russian authorities on charges of terrorism and two of his accomplices in Odessa on Feb. 4, but said nothing at the time about them being linked to an anti-Putin plot.
Its spokeswoman, Marina Ostapenko, said Monday the announcement in Moscow came only now because the Russian special service was conducting its own investigation. She confirmed the main suspect was involved in a plot to kill Putin, but didn't elaborate.
There was no immediate explanation for the different number of suspects cited by Russia and Ukraine.

An undated photograph taken from a Russian television report shows Ilya Pyanzin, who reportedly was conspiring to kill Vladimir Putin.
Putin's spokesman Dmitry Peskov confirmed the report to the ITAR-Tass news agency, but refused to make any further comment.
A laidback Yankee in trouble in Putin's court
Channel One said two of the alleged members of the group arrived in Ukraine from the United Arab Emirates via Turkey with instructions from Umarov, the top military leader for the Chechen rebels. One of them, a Chechen, was killed during the accidental explosion in Odessa and another one, Kazakhstan citizen Ilya Pyanzin, was wounded in the blast and arrested.
Pyanzin led the investigators to their contact in Odessa, Adam Osmayev, a Chechen who previously had lived in London and had been sought by Russia since 2007, the report said. The TV station showed footage of Osmayev's arrest in Odessa with black-clad special troops bursting in and a half-naked, bloodied Osmayev on his knees, his head bowed down.
Speaking to Channel One from custody in Ukraine, Osmayev described the group's mission: "Our goal was to go to Moscow and try to kill Prime Minister Putin ... Our deadline was after the Russian presidential election."
Both of Osmayev's hands were bandaged, and his face was covered in green dots from an antiseptic used to treat his cuts.
Russians rally for Putin -- and?2 days off work
Russian and Ukrainian special services wouldn't comment on the report.
The report is likely to boost support for Putin as he seeks his third term as president in an election Sunday.
But some Russians reacted to the news with skepticism, making clear on social network sites that they did not believe the report or suggesting the timing of the announcement was intended to attract sympathy for Putin before the election.
Opinion polls show Putin, a former KGB officer who crushed separatists during a war he launched in the Chechnya region in the North Caucasus before he became president, will easily win the election and reclaim the post he held from 2000 until 2008.
But he faces a growing opposition protest movement and wants to secure outright victory on Sunday, averting a runoff that might dent his authority.

This week I've had the pleasure of eating in the sister restaurants -?The Wayfarer and the Swan with Two Necks. On Tuesday there was a Food and Drink Festival working lunch with one of our sponsors. At one o'clock the restaurant was over two thirds full and everyone seemed to be enjoying their meals. I love The Wayfarer and am delighted it is continuing to do so well. We've dined here in the evenings before but this was my first time for lunch. The lunch menu is? most unusual and brings a totally different meaning to the word 'baguette'! I thoroughly recommend the crispy battered haddock variety!
Then last night Jan and I went over to the Swan with Two Necks. An almost full car park was the clue to the fact that they were busy. However that in no way diminished the wonderful experience we had. Initially we were shown to a table in a rather dim corner. Whilst in our younger days a table for two away from the crowd lit by a flickering candle may have had its attractions, at our age the eyes are slowly letting us down! We? need to be able to read the menu and see what we're eating! With no trouble at all, we were moved to another table and then spent a most enjoyable evening eating some outstanding food. I had the grilled mackerel starter (well cooked, firm and very tasty) followed by the belly pork main dish. Wow!
So of the two venues, do I have a favourite? No. They're both great restaurants serving great food at reasonable prices. But above all, it is so obvious that the management takes customer service seriously in both venues. All the staff were efficient and knowledgeable - and so friendly.?These restaurants are an asset to this area. If you haven't been yet, then I can recommend them both. You won't be disappointed.
And my diet? Well, last weekend?I totted up a weight loss of one and half stones since New Year's Day but I think I'll stay off the scales for a few days now!

Kombucha tea has become exceedingly trendy lately amongst the healthfully conscious, and stars such as Lindsay Lohan and Madonna have even been spotted sipping on the tea. But is it really the magical cure-all it claims to be?
The beverage has been around for more than 2,000 years; in fact, the ancient Chinese called Kombucha the ?Immortal Health Elixir.? Kombucha is a colony of bacteria and yeast, which is then added to sugar and tea and allowed to ferment in order to produce what is known as Kombucha tea. The tea is available commercially and can also be made at home.
Advocates of the brew claim that Kombucha detoxifies the body, increases energy, sharpens eyesight, aids joint recovery, improves skin elasticity, aids digestion and even prevents and fights cancer and arthritis. There is little published research done on the benefits.
As of yet there have not been any published case-controlled studies and no specific links have been established in relation to effect of central nervous system, liver, metabolic acidosis, and toxicity in general.
Several cases of harm have been reported, however, with symptoms including upset stomach, infections and allergic reactions. Some sources have even indirectly linked death to Kombucha.
Adverse reactions may be due to unsanitary fermentation conditions, leaching compounds from the fermentation vessels, or ?sickly? kombucha cultures that cannot acidify the brew. The acidity of the fermented drink is what prevents the growth of unwanted contaminants.
Since home-brewing facilities vary a great deal, the tea can easily become contaminated with harmful germs. Home-brewed tea should not be brewed in ceramic, lead crystal or painted containers, as the acidity of the beverage can cause it to absorb harmful elements from the container.
All in all, there is no solid evidence that Kombucha delivers on its health claims, and given that several cases of harm have been reported, it might be best to avoid it until further research has been done. Since potential health risks are unknown, anyone with an immune deficiency or other medical condition should consult physician before drinking the tea. Women who are pregnant or breast feeding should also avoid Kombucha.

PHOENIX (AP) ? A government informant whom defense attorneys have dubbed a "trailer park Mata Hari" didn't use sex to obtain evidence against two white supremacist brothers accused of bombing a city official in Arizona, a federal prosecutor said Tuesday.
Prosecutor Michael Morrissey told jurors that the hiring of the informant, identified in court records as Rebecca Williams, was "entirely permissible and necessary" to the investigation of identical twin brothers Dennis and Daniel Mahon.
The 61-year-old brothers are accused of sending a package bomb to Don Logan almost eight years ago to the day, on Feb. 26, 2004. Logan, 54, is black and was the diversity director for the Phoenix suburb of Scottsdale at the time.
Logan was hospitalized for three days after the bombing and needed four surgeries on his hand and arm. The bombing also injured a secretary.
The trial, which began Jan. 10, has been filled with drama as Williams took the stand for several days, and Logan himself described the bombing in detail for the jury.
Investigators chose the woman, who is a civilian, for her good looks and had her dress in revealing clothes and send the brothers racy photos to get them to open up to her. The "trailer park Mata Hari" nickname is a reference to the Dutch exotic dancer convicted of working as a spy for Germany during World War I.
Closing arguments began Tuesday morning, with Morrissey telling jurors that the evidence shows the Mahons were believers in "racial terrorism and violence as a way to accomplish their goals."
Morrissey replayed tapes that showed Dennis Mahon telling Williams how to make pipe bombs. In one recording, he tells her that he's had pipe bombs work in the past and that he's taught some white police officers how to make the explosives.
During the trial, jurors also heard recordings of the Mahons using racial epithets for black and Hispanic people and saying violence is the only answer for white men.
The brothers have pleaded not guilty to one count each of conspiracy to damage buildings and property by means of explosive, malicious damage of building by means of explosive, and distribution of information related to explosives.
Since the Mahons were arrested in June 2009 at their home in Davis Junction, Ill., their attorneys have unsuccessfully tried to get some or all of the charges against them dropped for various reasons, including allegations that the informant and federal investigators engaged in "outrageous conduct" that amounted to entrapment and coercion.
Defense attorneys also have argued that a more likely suspect in the bombing would be someone who worked in the city of Scottsdale and knew about its intraoffice mail, since the package was found in the city library and routed to Logan ? not sent in the mail.
The Mahons were living in the Phoenix area at the time of the bombing but left days afterward.
Prosecutors argued during the trial that the brothers belonged to a group called the White Aryan Resistance, an organization that encourages members to act as "lone wolves" and commit violence against non-whites and the government to get their message across.
They played for jurors hundreds of hours of video and audio surveillance of the brothers interacting with Williams.
In January 2005, Williams set up a government-provided trailer at a campground in Catoosa, Okla., where the brothers were staying after the bombing. Authorities put a Confederate flag in the window and had Williams act like a government separatist and a racist to get the brothers to trust her.
Over the years, the three mostly spoke over the phone, but they got together in person on several occasions. While apart, the informant sent the brothers several revealing photos of herself, including one of her from behind wearing chaps, a Confederate bikini bottom and a black leather jacket. Another photo showed her in a white bikini top with a grenade hanging between her breasts as she posed in front of a pickup truck and a swastika.
By all accounts the tactic worked.
Dennis Mahon opened up to her as their conversations were recorded. He bragged about bombing a Jewish community center, an Internal Revenue Service building, an immigration facility and an abortion clinic, according to court records. Those claims haven't been corroborated.
Williams gave hours of testimony in the case, telling jurors that the brothers fell so hard for her, Dennis Mahon said he wanted to marry her and father her children.
Dennis Mahon's attorney told jurors that her client exaggerated to Williams to impress her and get her into bed, while Daniel Mahon's attorney painted her client as a hard-working man who had no time or desire to plot a bombing.
While Logan was on the stand, he choked up as he recalled the terrifying moments when the bomb went off, describing how he frantically ran down a hallway before kneeling down, hearing a secretary scream, and looking down to see his hand and arm covered in blood.
He also re-enacted how he used scissors to cut one side of the package open, turned it 90 degrees to cut another side and then cut it down the middle before sticking his hand inside.
"I heard a pop that sounded like a gunshot and everything slowed down," said Logan, who said the next thing he remembers is feeling unbearable pain, the lights going out, the room filling with smoke, and debris falling from the ceiling.
The same day in court, prosecutors played for jurors a voicemail that Dennis Mahon left at the diversity office a few months before the bombing.
"The white Aryan resistance is growing in Scottsdale," Dennis Mahon said angrily in the recording. "There's a few white people who are standing up."

AP - RIO DE JANEIRO — The Carnival band kicked off according to tradition: Dozens of percussionists struck their bass drums, sending a low rumble like a sustained thunderclap echoing among the skyscrapers of Rio's downtown. Tambourines answered lightly, tinkling over the drummers' resounding call. Dancers responded with a roar, throwing up their hands, feet leaping to the frenetic beat.
[unable to retrieve full-text content]HealthDay - MONDAY, Feb. 20 (HealthDay News) -- Certain complications during pregnancy appear to raise the mother's risk of cardiovascular disease during middle age, a new study has found.
Objective: To study the expression of cyclooxygenase-2 (cox-2) and angiopoietin-2 (Ang-2) in colorectal cancer tissue, tissue adjacent to the cancer, normal tissue and the relationship between these expressions and the clinic pathological parameters of colorectal cancer. And to investigate the effect on cell growth, expression of VEGF and Angiopointin-2 protein in HT-29 cell line with the treatment of Meloxicam, a selected NSAIDs.Methods: Immunohistochemical SP technique method was used to detect the expression of cox-2 and Ang-2 protein in 45 colon cancer tissue, 45 tissue adjacent to the cancer and 15 normal tissue. Then cultured HT-29 cells were treated by Meloxicam with various concentration and time. Cell Counting Kit-8 (CCK-8) detected the proliferation of HT-29. The levels of VEGF and Ang-2 protein in supernatants were determined by enzyme-linked immunosorbent assay (ELISA). The expression of VEGF and Ang-2 mRNA in cultured HT-29 were determined by real-time quantitative reverse- transcription-polymerase chain reaction. Results: cox-2 and Ang-2 were expressed in tumor tissue and tissue adjacent to tumor, were not expressed in normal tissue. In 45 colon cancer tissue, the positive expression ratio of cox-2 and Ang-2 were 80.0%, 66.7%respectively; in 45 tissue adjacent to the cancer, the positive expression rates of cox-2 and Ang-2 were 35.6%, 11.1%. There was a close correlationship between the expressions of cox-2 and Ang-2 with several clinic pathological factors including lymph node metastasis and Dukes stage. And there was a close correlation between the expression of cox-2 and Ang-2. And there was a cytotoxicity in cell proliferation of HT-29 with Meloxicam treatment in different concentration and time, a concentration-effect and time- effect relationship was showed to some extent. Cell cycle analysis showed that the cells were mainly blocked in G0/G1phase.The VEGF and Ang-2 protein level in supernatants was reduced gradually with the concentration or time increasing; there was a dependence on the concentration and action time. The expression of cox-2?VEGF and Ang-2mRNA was reduced gradually with the concentration increasing, and also there was a dependence on the concentration. Conclusion: cox-2 and Ang-2 play an important role in the happen and development of colon cancer..Meloxicam can reduce the expression of VEGF and Ang-2 at the level of protein and mRNA in colon carcinoma cell line. Application of specific inhibitor of cox-2 in clinic is significative.

AP - The growl came first, low and throaty, piercing the darkness that had fallen across the remote Australian desert. A baby's cry followed, then abruptly went silent. Inside the tent, the infant girl had vanished. Outside, her mother was screaming: "The dingo's got my baby!"
As Havard Business School professor Josh Lerner thanked the audience to conclude his interview of NEA Chairman and Co-founder Dick Kramlich, the keynote event this morning at this year?s?HBS Venture Capital and Private Equity Conference, Kramlich interrupted him. Speaking to a room of hundreds of business school students, Kramlich took care to reemphasize the point he wanted to drive home: ?The venture capital business is the largest single creator of jobs in the U.S.,? he said. And in his view it is alive and well.
Kramlich began his venture career in 1969, nine years before he founded NEA, in the period that he describes as beginning with Intel and ending with Apple. That period was a difficult one for the U.S. economically, with an oil crisis and years of ?stagflation? ? high inflation coupled with slow economic growth ? but Kramlich called the climate ?a great lesson? in how to succeed in venture capital.
As he looks out at today?s business climate, Kramlich sees huge potential in China, but cautioned his audience that taking advantage of the country?s opportunity requires serious commitment.
?I think they?re the sign of spring in the capital markets,? he said, speaking on China. But ?flying in and out doesn?t work.??The Chinese market can be prone to ?overheating? with so many investors looking at that market, so VCs have to ?have a longer term?point?of view.?
Asked about India, Kramlich was more tempered, citing greater complexity and layers of bureaucracy.
But for the venture industry overall, he was optimistic.
?We?ve gotten better practices,? he said. ?We?ve improved on our process over time.? Asked about sectors he favored, Kramlich compared venture investing to a buffet rather than a one course meal, saying that global funds like NEA would do well to look broadly across sectors.
But he doesn?t deny that the venture business is changing. For instance, he considers the rise of angel groups and ?super angels? as a positive development.
?We actually regard ourselves as partners to the angels,? he said. ?My religion is ?whatever is good for the entrepreneur is good for us.??

A good subject is not the only requirement for a good or award winning photograph. There are various other elements that go in the capturing of a masterpiece. A majority of the photographers tend to concentrate on a few elements and ignore a few others that are quite crucial in getting the best shots that can be showcased by an individual in order to promote his or her photography skills. The choice of the subject is crucial although it is not the final element that will decide the quality of the image that you are going to click.?
?
Composition of the image forms an important part of the elements that go into the making of a good photograph. A good subject lacking good composition is nothing less than an expensive piece of antique lying in a scrap dealer?s shop. Thus composition is of utmost importance once you have decided to go pro in terms of clicking images. Make sure the subject you have chosen is highlighted in photographs. If you are clicking a rose with dew drops on it the onlooker's eyes must see the dew and the petals on which the dew is instead of the other parts of the flower.?
?
You are not clicking a photograph, you are reading a story to your audience, a story that is self captivating and needs no explanation to people who have a look at it. The image needs to explain the various moods and feelings that have been captured. If its a joyful image showing happiness on the face of the subjects the reason for that happiness should be captured behind the subject. If you are clicking a content cow showing expressions of satisfaction a green meadow in the background will be the perfect composition that is going to share the story of the cow's satisfaction and the prosperous surroundings that she is in.?
?
Using the rule of thirds where you draw three horizontal and three vertical lines and place the subject at the intersection of these lines is quite effective and would surely bring out the beauty of the subject in the final image that is presented before the audience. Make sure that the photos that are clicked by you are well exposed. Although you may change the light effects later on with the help of software but natural exposure is something that will evoke the best responses fro your images.?
?
Make sure your photographs evoke feelings among your audience; a child's smile should be able to stir enough love and affection inside a human being. A war scene should be able to evoke the feelings of horror and sufferings. If you are able to take your audience on a roller coaster ride of mood swings then think that you are clicking a right mix of images with a right proportion of all ingredients that go in the making of a good stock photo.

The home enterprise business is expanding continuously and analysis advised that revenue generated from home-based enterprise will improve by 17% yearly through the yr 2008.
A person important characteristics of the home base world-wide-web enterprise is it requires an incredibly Lower Investment Price but Very Higher Investment RETURNS after it?s established and jogging. Compare with other sorts of expense like stocks, qualities and classic businesses, if the expense gone badly, you may finish up shedding the principal as well as the revenue obtained.
Furthermore, home centered enterprise is much more flexible and it could be in any type. The doing work hour isn?t going to required be from nine to 5 every day, it is possible to begin work at any time as well as any where by provided that you?ve access to a laptop or computer. The enterprise may be your know-how advertising, i.e. sell an ebook about the best way to fishing; or physical item advertising, by way of example buying and selling on ebay; or doing surveys for corporation; or be able to write ads for firms to promote their item and generate commissions.
One of many most popular home centered businesses is Network marketing or Multi-level Advertising and marketing(Multi level marketing). For above half of century, multi-level marketing has grown rather a little. Most network marketers earning from the number of hundred bucks to a number of thousand bucks per month. But importantly they liked the freedom of doing work for on their own.
The concept of multi-level marketing is simple. An organization can offer their solutions and services at discount prices to you personally then by advertising the products/services to other folks at a total selling price, that you are earning the main difference because the commission.
Thus world-wide-web home base enterprise is your Ideal wager in direction of fiscal protection. The moment your home centered enterprise is on course, you?ll be experiencing your fiscal independence by acquiring two earning resources. A person out of your total time position along with the 2nd one particular coming out of your home centered world-wide-web enterprise. The majority of all, there may be large probable in home based business earnings to OUTGROWN your total time earnings.

The hashtags #ThingsAspirinCures and #backinmyday may be the only good things to come out of the ridiculous comments Foster Friess made on Thursday.
The Rick Santorum backer and super PAC contributor shared his thoughts on contraception (and Bayer aspirin, for that matter) with the MSNBC anchor Andrea Mitchell on her show.
"This contraceptive thing, my gosh, it's such [sic] inexpensive," Friess said on the show. "Back in my days, they used Bayer Aspirin for contraceptives. The gals put it between their knees, and it wasn't that costly."
While Mitchell was at a loss for words, Twitter nearly exploded with reactions and "Foster Friess" was soon a top trending topic in the U.S.

Technical writing is a very vital part of communication. For using certain goods and services we may face some technical hurdles. Technical communication becomes essential in such cases. Technical assistance is provided by writers who are expert in this field. Technical writing or content cannot be a limited to articles explaining the technical process. These articles need to be supported with diagrams, self-explanatory charts, multimedia guides etc. Hence technical writing services will include writing, providing illustrations, designing, and editing the final piece of information. Technical writing involves making user manuals, administration charts, program and installation guides, and manuals for software applications. Good technical writing is well appreciated only when it presents complex information in clear and concise way.
Such writing is necessary for computer programming , electronic, scientific research and financial industries.
For technical writing, studying the audience to whom the article is targeted is very important. The writer has to have sound knowledge about the product, the customer or the reader and the reason behind developing the technical document for the reader. Another basic feature that has to be kept in mind is the cost involved. The technical writer needs to tailor the content to readers belonging to various background. The writer has to excel here by providing information in such a way that it is useful for all. For instance, a technical writer may be making a instruction guide for installing a software. He has to be cautious not to use any technical jargons and even if he does the meaning should follow in a sub bracket.
The writer needs to be careful about the choice of vocabulary and the style of writing. The articles should be also appropriate in length and include some case studies to make it meaningful. Demonstration is the ideal way to deal with such technical matters. An illustrative manual is always preferred to a page full of words.
Another specialized area of technical writing is financial writing. Financial documents are full of data and figures and all of them together convey some specific meaning. Interpreting them is the task of subject matter expert. These experts are stalwarts in commercial domain and could involve in writing simpler articles to convey the crux of the financial documents along with its implication. They could advise and guide other writers in preparing some lucid articles that makes greater sense to the wider audience. Business writing services are provided by professionals having good knowledge of the industry and the respective companies.
Jenny Stewart hired some people who specialised in technical writing to make manuals and guide books for their products and softwares. These handy manuals helped customers in using the products with greater ease. Reputation of the products have been enhanced.

Harvard's Wyss Institute develops DNA nanorobot to trigger targeted therapeutic responsesPublic release date: 16-Feb-2012 [ | E-mail | Share ]

Novel technology could potentially seek out cancer cells and cause them to self-destruct
Researchers at the Wyss Institute for Biologically Inspired Engineering at Harvard University have developed a robotic device made from DNA that could potentially seek out specific cell targets within a complex mixture of cell types and deliver important molecular instructions, such as telling cancer cells to self-destruct. Inspired by the mechanics of the body's own immune system, the technology might one day be used to program immune responses to treat various diseases. The research findings appear today in Science.
Using the DNA origami method, in which complex three-dimensional shapes and objects are constructed by folding strands of DNA, Shawn Douglas, Ph.D., a Wyss Technology Development Fellow, and Ido Bachelet, Ph.D., a former Wyss Postdoctoral Fellow who is now an Assistant Professor in the Faculty of Life Sciences and the Nano-Center at Bar-Ilan University in Israel, created a nanosized robot in the form of an open barrel whose two halves are connected by a hinge. The DNA barrel, which acts as a container, is held shut by special DNA latches that can recognize and seek out combinations of cell-surface proteins, including disease markers. When the latches find their targets, they reconfigure, causing the two halves of the barrel to swing open and expose its contents, or payload. The container can hold various types of payloads, including specific molecules with encoded instructions that can interact with specific cell surface signaling receptors.
Douglas and Bachelet used this system to deliver instructions, which were encoded in antibody fragments, to two different types of cancer cellsleukemia and lymphoma. In each case, the message to the cell was to activate its "suicide switch"a standard feature that allows aging or abnormal cells to be eliminated. And since leukemia and lymphoma cells speak different languages, the messages were written in different antibody combinations.
This programmable nanotherapeutic approach was modeled on the body's own immune system in which white blood cells patrol the bloodstream for any signs of trouble. These infection fighters are able to hone in on specific cells in distress, bind to them, and transmit comprehensible signals to them to self-destruct. The DNA nanorobot emulates this level of specificity through the use of modular components in which different hinges and molecular messages can be switched in and out of the underlying delivery system, much as different engines and tires can be placed on the same chassis. The programmable power of this type of modularity means the system has the potential to one day be used to treat a variety of diseases.
"We can finally integrate sensing and logical computing functions via complex, yet predictable, nanostructuressome of the first hybrids of structural DNA, antibodies, aptamers and metal atomic clustersaimed at useful, very specific targeting of human cancers and T-cells," said George Church, Ph.D., a Wyss core faculty member and Professor of Genetics at Harvard Medical School, who is Principal Investigator on the project.
Because DNA is a natural biocompatible and biodegradable material, DNA nanotechnology is widely recognized for its potential as a delivery mechanism for drugs and molecular signals. But there have been significant challenges to its implementation, such as what type of structure to create; how to open, close, and reopen that structure to insert, transport, and deliver a payload; and how to program this type of nanoscale robot.
By combining several novel elements for the first time, the new system represents a significant advance in overcoming these implementation obstacles. For instance, because the barrel-shaped structure has no top or bottom lids, the payloads can be loaded from the side in a single stepwithout having to open the structure first and then reclose it. Also, while other systems use release mechanisms that respond to DNA or RNA, the novel mechanism used here responds to proteins, which are more commonly found on cell surfaces and are largely responsible for transmembrane signaling in cells. Finally, this is the first DNA-origami-based system that uses antibody fragments to convey molecular messagesa feature that offers a controlled and programmable way to replicate an immune response or develop new types of targeted therapies.
"This work represents a major breakthrough in the field of nanobiotechnology as it demonstrates the ability to leverage recent advances in the field of DNA origami pioneered by researchers around the world, including the Wyss Institute's own William Shih, to meet a real-world challenge, namely killing cancer cells with high specificity," said Wyss Institute Founding Director, Donald Ingber, M.D., Ph.D. Ingber is also the Judah Folkman Professor of Vascular Biology at Harvard Medical School and the Vascular Biology Program at Children's Hospital Boston, and Professor of Bioengineering at Harvard's School of Engineering and Applied Sciences. "This focus on translating technologies from the laboratory into transformative products and therapies is what the Wyss Institute is all about."
